WEBPharmacology. Tapentadol is a unique, centrally acting opioid of the aromatic hydrocarbon benzenoid class 29 (Figure). Tapentadol is a nonracemic molecule and, unlike tramadol, is not a prodrug. Its analgesic effect is produced through moderate opioid receptor agonism and norepinephrine reuptake inhibition.
